What is Windows Defender? 

Last night I was installing  IE9 on my PC, during which it switched "OFF" Microsoft Security Essentials And Windows Defender. So, I switched "ON" MSE but, should I switch on Windows Defender too?

I tried to switch "ON" Windows Defender, but it was talking a lot of time, what could be the reason?

What is the basic difference between MSE and WINDOWS DEFENDER?

Windows Defender is an alternate to MSE, which come as pre-installed update to your Windows. But when you install MSE, it automatically turns off the Windows Defender to prevent the conflict between the two. You dont need to turn on windows defender, and in some cases you be able to do even if you try. So dont be afraid if its not working. MSE will take care of all your threats.

What about time consumption for switching on " WINDOWS DEFENDER"? Does it have something to do with "MSE"?

Windows Defender is an older anti-spyware tool developed by Microsoft. I comes pre-installed with Windows Vista and 7.

Microsoft Security Essentials (MSE) is the latest Anti-malware and anti-spyware tool from Microsoft and its better than Windows Defender. As UG said, it automatically turns Windows Defender off, so you don't need to enable it again as its useless and will also consume system resources.

So forget about Windows Defender and keep using MSE.
